چکیده

Abstract Genotyping by sequencing (GBS) and single nucleotide polymorphism (SNP) chip technologies are the primary SNP genotyping used today. However, these have some drawbacks that limit their usefulness in analysis. We developed a robust protocol called multiplexed (GMS) using markers, providing informative genotypic data with greater flexibility. The genotypes derived from direct sequence reads reduce ambiguity genetic advantages of this include: (1) This PCR-based generates information markers interest provides more streamlined accurate analysis process, multiplexing hundreds into run. (2) marker sets easily customized to species can readily be changed. In study we taken GMS wheat adapted it barley. identified 577 work well adequate genome coverage for genomic selection tag 267 QTL’s genes interest. Good an read depth at least 5 amplicons reliably present across population.

Genotyping-by-Sequencing in Plants

The advent of next-generation DNA sequencing (NGS) technologies has led to the development of rapid genome-wide Single Nucleotide Polymorphism (SNP) detection applications in various plant species. Recent improvements in sequencing throughput combined with an overall decrease in costs per gigabase of sequence is allowing NGS to be applied to not only the evaluation of small subsets of parental ...

Multiplexed DNA sequencing-by-synthesis.

We report a new DNA sequencing-by-synthesis method in which the sequences of DNA templates, hybridized to a surface-immobilized array of DNA primers, are determined by sensing the number of nucleotides by which the primers in each array spot are extended in sequential DNA polymerase-catalyzed nucleotide incorporation reactions, each with a single fluorescein-labeled deoxyribonucleoside triphosp...

Ordered catenation of sequence-tagged sites and multiplexed SNP genotyping by sequencing.

We describe a method for the efficient genotyping of SNPs, involving sequencing of ordered and catenated sequence-tagged sites (OCS). In OCS, short genomic segments, each containing an SNP, are amplified by PCR using primers that carry specially designed extra nucleotides at their 5'-ends. Amplification products are then combined and converted to a concatamer in a defined order by a second roun...
